What is the Extraction process of Rosemary?
xtraction process:
1. Supercritical CO2 fluid extraction: Put 160.0g of rosemary powder, which was crushed evenly and sifted, into a 1L extraction kettle. With oil yield as the index, an orthogonal experiment of extraction time, extraction pressure and extraction temperature was designed.Repeat twice and take the arithmetic mean.The optimum extraction conditions were: extraction pressure of 20.0MPa, extraction temperature of 40.0℃, extraction time of 4.0h, and oil yield of 4.40%.
2. The water vapor distillation: will shatter evenly after sifting rosemary powder in 500 ml of 20.0 g round bottom flask, to yield efficiency as an index, the design of material particle size, liquid ratio and extraction time of 3 factors 3 levels orthogonal test, according to the law of the People's Republic of China pharmacopoeia (2000 edition) of volatile oil determination method in appendix XD, repeat 3 times, the results take arithmetic average.The optimal technological conditions were as follows: the particle size of the material was 0.20mm, the liquid-solid ratio (V/W) was 4:1, the extraction time was 4.0h, and the oil yield reached 1.40% at this time.
Content analysis: 10ml of the sample was accurately measured according to the ester determination method (OT18).The equivalent factor (e) in the calculation is 98.15.The ester content in terms of borneol acetate was obtained.The total borneol content was determined by total alcohol content determination method (OT-5).During saponification, 5ml of dry acetylated oil should be accurately measured.The percentage of Chemicalbook borneol (calculated by the following formula: X= 7.712a (1-0.0021e)/(B-0.021a) in formula A -- the difference between the consumption of 0.5mol/L hydrochloric acid volume (mL) of the titration sample solution and the consumption of blank test;B The quality of one acetylated oil, g;E - Ester content in bornyl acetate (C12H20O2), %(see above).
